The composition of the calcified matrix of long bone from male Sprague-Dawley rats on different diets was analysed by HPLC and analyzed by SDS-PAGE and image analysis. The content of apatite, as analysed by x-ray diffraction, was 0.30 and 0.34 g/cm3 in newly formed bone from rats on diets containing 1.25 g/100 g protein (control diet) and 0.08 g/100 g protein (diet without calcium), respectively. The possible involvement of carbohydrates in the mineral matrix of newly formed bone was demonstrated by SDS-PAGE and a zymographic technique. The carbohydrate moiety was released by proteolytic treatment of the acid-soluble matrix and it was positive for periodic acid Schiff, indicating the presence of glycosaminoglycan structures.
