Glutamate release from the dorsal spinal cord via Ca2+-dependent release is differentially regulated in the nociceptive C- and H-fibres.
This study was designed to examine whether the release of glutamate from dorsal spinal cord in response to noxious and non-noxious stimulation was regulated by different Ca2+ sources and/or Ca2+ transporters. Transmitter release was measured in the superfusate from dorsal roots of spinal cords from adult, male rats using the microdialysis technique. A significant increase in the glutamate concentration was observed after noxious stimulation with a 100% increase above basal release in C-fibres and a 107% increase in H-fibres. The non-noxious stimulation of the muscles (hypertonic saline) resulted in a 44% increase in C-fibre glutamate release, and a 110% increase in H-fibre release. The glutamate release in the dorsal spinal cord was significantly inhibited by kynurenic acid and inhibited to a lesser extent by specific inhibitors of voltage-gated Ca2+ channels, the fast inactivation gate of voltage-gated Ca2+ channels and the ryanodine receptor type 1 (RyR1). The increase in H-fibre glutamate release in response to noxious stimulation was significantly inhibited by glutamate (Glu) uptake blockers, an indication of the presence of high-affinity uptake. In contrast, the noxious stimulation of C-fibres in the presence of Glu uptake inhibitors resulted in a significant increase in the release. The increase in C-fibre glutamate release in response to noxious stimulation was inhibited by bath application of the N-methyl-d-aspartate receptor antagonist APV and the non-N-methyl-d-aspartate receptor antagonist CNQX. The increase in glutamate release in response to noxious stimulation was significantly inhibited by several types of Ca2+-dependent K+ channels, including Kv1.1, Kv1.2 and CaV1.2 (L-type). The noxious stimulation-induced increase in H-fibre glutamate release was significantly inhibited by bath application of the Ca2+
